Adeno-associated viruses (AAVs) are the main viral vectors for gene therapy and as they continue to be evaluated as therapeutic delivery platforms, it is critical to ensure that all the critical quality attributes (CQAs) of the therapeutic product are maintained. Viral capsid protein characterization yields several challenges. While traditionally, SDS-PAGE is used to establish the molecular weight of the capsid proteins, it only provides an approximate molecular weight. Mass spectrometry is a promising method to overcome these challenges and determine CQAs of the capsid proteins.
