The enzyme horseradish peroxidase (HRP) can be attached to your protein or antibody to create signal-generating molecules. Sugar groups on native HRP are oxidized with periodate to produce amine-reactive aldehydes. The activated peroxidase spontaneously and efficiently crosslinks with primary amines on the antibody or other proteins. This method directs the coupling location to the Fc portion of the antibody molecule and is more effective than other amine-reactive chemistries, such as glutaraldehyde coupling, which often causes polymerization and conjugate inactivation.
